METHOD OF DIFFERENTIATION OF PLURIPOTENT STEM CELLS TO DEFINITIVE ENDODERM CELLS

Method for inducing differentiation of pluripotent stem cells to cells which are precursors of tissues derived from endoderm. The cells obtained may be used for regenerative medicine and somatic cell therapies.

Stem cells are a promising source for the development of new cell therapies in the context of regenerative medicine. The application of these cells include replacement of damaged cells or tissues, treatment of degenerative diseases such as diabetes, Parkinson or Alzheimer, medular lessions, brands, osteoporosis, etc.

There are some methods in the art directed to the induction of differentiation of pluripotent or multipotent stem cells, e.g. using hyaluronic acid, a collagene matrix, Activin A, etc. In particular, the exclusion of LIF and the presence of VPA or NO has also been described. However, a method combining VPA and NO in the absence of LIF has never been disclosed.

The present research group has developed a method of differentiation of pluripotent stem cells involving culture of the cells in the presence of VPA and NO and absence of LIF. The method offered requires shorter time than those known in the prior art.
