Del Priore, M.D., Ph.D. Problem or Unmet Need: The medical devices industry is one of the most vital, dynamic and fastest-growing sectors in the healthcare market; however its full potential is limited in part by lack of patient confidence due to device defects, infections and blood clots. One key method to reduce blood clots (and consequently improve patient confidence) is to prevent the cell-cell adhesion that is often induced by medical devices by inhibiting adhesion molecules such as beta-1 integrin. Details of the Invention: This invention identifies CD46, a beta-1 integrin interacting membrane protein expressed on all nucleated cells, as a novel protein in cell adhesion. In the presence of a CD46 inhibitor, cell adhesion to other cells or substrate is reduced to levels comparable to that of beta-1 integrin inhibition. This invention provides a medical device for use in inhibiting adhesion of cells, wherein the medical device has a coating comprising an inhibitor of CD46. Applications: • A therapeutic target for cell-cell or cell-substrate adhesion modulation that can be coupled to medical device implantation • Therapeutic applications include, but are not limited to: o Adhesion inhibitor coating of arterial stents for treatment of heart disease o Promotion of re-epithelialization of the internal lumen of the stent o Adhesion inhibition in ophthalmic medical device implantation o Inhibition of cell adhesion involved in the neoplastic growth of some cancers Advantages: • Can be targeted via a device (e.g. implantable slow-release device) or by direct delivery • Multiple forms of cell-adhesion modulation: o CD46 inhibition to prevent blood clotting o CD46 activation to promote wound healing • A novel and alternative target for cell-adhesion modulation Patent Status: Patent Pending (
