A new method for prevention and/or cure of type 1 diabetes based on: 1° the intrathymic expression of IGF2 (Insulin-like growth factor 2) as the dominant gene of the insulin family; 2° the defect of Igf2 expression in the thymus of animal model of type 1 diabetes; and 3° the regulatory/tolerogenic profile of cytokine secretion induced by DQ8 presentation of IGF-2 B11-25 in cultures of PBMCs isolated from DQ8+ type 1 diabetic adolescents. Potential applications : Design of a novel type of tolerogenic (negative) vaccine to inhibit the development of the autoimmune response selective of insulin-secreting islet ß cells.
