Novel diurea lead compounds that block intracellular signal transduction and inhibit interleukin-2 production and other pro-inflammatory cytokines in T-cells, and promote activation-induced cell death (apoptosis) for treatment of inflammatory diseases. These compounds were discovered in a screening process for novel signal transduction inhibitors with the ability to block the production of IL-2 and other pro- inflammatory cytokines in activated T-cells. The aim is to develop a new anti-inflammatory drug for the treatment of conditions involving chronic inflammation such as rheumatoid arthritis (RA) and inflammatory bowel disease (IBD). The diurea compounds are potent inhibitors of IL-2 production in human CD4+ T-cells. They display IC50 values in the range of 1 micro M in cell-based assays. Proof of concept in vivo has been obtained demonstrating significant inhibition of IL-2 production induced by superantigen.
A priority patent application has been filed 2004-02- 04, which claims protection for novel diurea compounds, that inhibit the production of pro-inflammatory cytokines and/or induce apoptosis in activated T-cells, use of such a compound as a medicament, a pharmaceutical composition comprising said compounds and a method of treatment. At the same date a provisional US patent application was filed. Status: Pending applications. Expected expiry date: 2025.
